Effect of Chemical Sterilization on the Mechanical Properties of Human Cortical Bone Allografts

چکیده
Introduction Musculoskeletal allografts are increasingly being utilized in a variety of orthopedic procedures. However, they pose the serious risk of viral or bacterial disease transmission as highlighted by the 2001 death of a young man from Clostridium sordellii sepsis after receiving a contaminated allograft. Gamma irradiation has been widely used to minimize this risk, but has been shown to weaken musculoskeletal allografts. Therefore, the need exists for sterilization techniques that do not diminish allograft biomechanical properties. BioCleanseTM (Regeneration Technologies Inc., Alachua, FL) is a fully-automated, low-temperature, chemical sterilization process that removes endogenous donor material such as blood and lipids, and has been validated to consistently eliminate bacteria, bacterial spores, fungi, and viruses. The objective of this study was to evaluate the effects of this sterilization technique on bone strength.

Biomechanical Integrity of Human Allograft Bone After Sterilization
Musculoskeletal allografts have been used extensively for a variety of orthopaedic procedures. Allograft does have the potential, however, to transmit a variety of pathogens. Two current sterilization methods widely used in the tissue banking industry are ethylene oxide (ETO) treatment and gamma irradiation. ETO has been shown to adversely affect the biologic incorporation of allograft tissue. ...

versus ethylene oxide (ETO) sterilization on the mechanical strength of cortical bone grafts. Tibias were collected from cadavers of mature goats. Sixty test specimens were randomized into four groups : fresh (no processing), frozen (freezing at –70°C), gamma-irradiated, and ETO-sterilized specimens. Torsion, three-point bending, and compression testing were separately performed with a material...
متن کاملReducing the radiation sterilization dose improves mechanical and biological quality while retaining sterility assurance levels of bone allografts.
BACKGROUND Bone allografts carry a risk of infection, so terminal sterilization by gamma irradiation at 25kGy is recommended; but is deleterious to bone quality. Contemporary bone banking significantly reduces initial allograft bioburden, questioning the need to sterilize at 25kGy. METHODS We inoculated allograft bone with Staphylococcus epidermidis and Bacillus pumilus, then exposed them to ...
متن کاملWhich is the best method of sterilization of tumour bone for reimplantation? a biomechanical and histopathological study
INTRODUCTION Sterilization and re-usage of tumour bone for reconstruction after tumour resection is now gaining popularity in the East. This recycle tumour bone needs to be sterilized in order to eradicate the tumour cells before re-implantation for limb salvage procedures. The effect of some of these treatments on the integrity and sterility of the bone after treatment has been published but t...
متن کاملEffect of Gamma Irradiation on the Mechanical Properties of Morselised Allograft
INTRODUCTION: Impaction grafting is a common procedure in the reconstruction of cavity defects. Concerns regarding sterility of the graft material may require gamma irradiation prior to implantation. The grafts’ mechanical properties play an important role in surgical handling and in-vivo response. Gamma irradiation adversely affects the mechanical properties in a dose dependent fashion in cort...
